font-weight: bold;\">colite ulcerosa<\/span>\u00a0e la\u00a0<span style=\"color: #cfaf7a; font-weight: bold;\">colite ulcerosa<\/span>, grazie alla sua capacit\u00e0 di danneggiare selettivamente l\u2019epitelio intestinale e compromettere l\u2019integrit\u00e0 della barriera mucosale del colon distale e rettale. Questo danno induce manifestazioni cliniche coerenti con le patologie umane: <span style=\"color: #cfaf7a; font-weight: bold;\">calo ponderale, diarrea e sangue occulto nelle feci<\/span>, rendendolo uno strumento validato per studi traslazionali nelle IBD<b>. <span style=\"color: #cfaf7a; font-weight: bold;\">Viene utilizzato per testare l\u2019efficacia terapeutica di nuovi farmaci.<\/span><\/b><\/span><\/p><p><span style=\"font-weight: 400;\"><span style=\"color: #cfaf7a; font-weight: bold;\">\ud83d\udd0d\u00a0 COME FUNZIONA IL MODELLO?<b> <\/b><\/span>La <span style=\"color: #cfaf7a; font-weight: bold;\">fase acuta<\/span> <\/span><span style=\"font-size: 16px; font-weight: 400;\">\u00e8 caratterizzata da una marcata attivazione di <\/span><span style=\"color: #cfaf7a; font-weight: bold;\">colite neutrofili e macrofagi<\/span><span style=\"font-size: 16px; font-weight: 400;\">. Nelle <span style=\"font-weight: 400;\"><span style=\"color: #cfaf7a; font-weight: bold;\">fasi croniche<\/span><\/span>, invece, si osserva un <span style=\"font-weight: 400;\"><span style=\"color: #cfaf7a; font-weight: bold;\">maggiore coinvolgimento dei linfociti<\/span><\/span>, in linea con le dinamiche immunitarie che si riscontrano nei pazienti affetti da IBD. <span style=\"font-weight: 400;\"><span style=\"color: #cfaf7a; font-weight: bold;\">Il modello \u00e8 ben validato e garantisce un\u2019elevata riproducibilit\u00e0 acqua da bere) di DSS al 2,5-3% (massa molecolare 40 kDa), caratterizzato da 7 giorni di esposizione al DSS seguiti da 3-7 giorni di acqua depurata.<\/span><\/p><p><span style=\"font-weight: 400;\">Nel modello di <span style=\"color: #cfaf7a; font-weight: bold;\"><b>colite cronica<\/b><\/span><\/span><b>\u00a0<\/b><span style=\"font-weight: 400;\">i topi C57\/Bl6 di 8-12 settimane vengono sottoposti a 3 cicli orali di DSS al 2%, caratterizzato da 7 giorni di esposizione al DSS seguiti da 7 giorni di acqua depurata.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div style=\"font-weight: 400;\">L\u2019apparato sperimentale per l\u2019endoscopia, denominato \u201cColoview system\u201d, \u00e8 composto da un endoscopio in miniatura (diametro esterno di 1,9 mm), una fonte di luce allo xeno, una telecamera a triplo chip e una pompa d\u2019aria (Karl Storz, Tuttlingen, Germania).<\/span><\/p><p><span style=\"font-weight: 400;\">La procedura endoscopica viene visualizzata su un monitor a colori e un punteggio endoscopico viene applicato in base ai segienti parametri: <\/span><span style=\"font-weight: 400;\">\u00a0traslucenza del colon (0\u20133 punti), granulosit\u00e0 della mucosa (0\u20133 punti), morfologia del pattern vascolare (0\u20134 punti) e presenza di fibrina (0\u20134 punti). <\/span><\/p><p><span style=\"font-weight: 400;\">Il punteggio cumulativo varia da 0 (assenza di segni di infiammazione) a 14 (segni endoscopici di infiammazione molto grave).(<\/span><span style=\"font-weight: Un patologo esperto, in cieco rispetto al trattamento, valuta l\u2019infiammazione e i processi di riparazione epiteliale per assegnare un punteggio istologico, secondo quanto descritto da Rachmilewitz\u00b9 o Dieleman\u00b2 et al. Le immagini rappresentative mostrano che nei topi sani (es. Healthy + Vehicle) gli strati tissutali (mucosa, muscularis mucosae e sottomucosa) risultano ben strutturati. Nei topi con colite severa indotta da DSS (es. DSS + Vehicle), la stratificazione degli strati non \u00e8 pi\u00f9 mantenuta: un massiccio infiltrato infiammatorio (cerchi rossi) occupa completamente sia la mucosa che la sottomucosa, e le cripte risultano gravemente danneggiate (cerchio giallo) nella maggior parte del colon distale.
